Comparative Evaluation of Cytotoxicity of a Novel Neem-Lemongrass Formulation and 3% Sodium Hypochlorite as a Root Canal Irrigant - An In Vitro Study

Background: Modern endodontics greatly emphasizes the use of root canal irrigants. Since the past decade, there have been numerous in vitro and in vivo studies done using herbal based endodontic irrigants. The main reason for this is the comparatively lower toxicity and side effects of herbal irrigants.
Aim: The aim of this study was to compare and analyze the cytotoxicity of a novel Neem-Lemongrass formulation with 3 % sodium hypochlorite as a potential root canal irrigant.
Materials and Methods: Ethanolic extract of the Neem-Lemongrass formulation was prepared. For cell viability assay, L929 cells cultured on EMEM in a 96-well tissue culture plate was used. The cell viability of different concentrations of the herbal irrigant (6.25, 12.5, 25.0, 50.0 and 100 µg/ml) and 3% sodium hypochlorite was assessed by MTT assay. 20 µl of MTT was added to each well. The absorbance was read on an ELISA microplate reader at 570 nm. Statistical analysis was performed using ANOVA.
Results: Significantly higher absorbance values were obtained for the herbal irrigant as compared to 3% NaOCl (P<0.05). Highest cell viability was for the negative control (100%), followed by 6.25µg/ml (94%), 12.5µg/ml (89.49%), 25µg/ml (73.9%), 50µg/ml (62.18%), 100µg/ml (50.94%) of the Neem-Lemongrass irrigant and finally 3% NaOCl (28.87%) (p<0.05). The 50% inhibitory concentration (IC50) of the Neem-Lemongrass irrigant was approximately 100µg/ml.
Conclusion: In conclusion, the cytotoxicity of the Neem-Lemongrass formulation at a concentration of 100µg/ml was much lower than 3% sodium hypochlorite and can be considered as a potential endodontic irrigant.
