Antimicrobial Efficacy of a Novel Neem-Lemongrass Formulation as a Root Canal Irrigant against Enterococcus Faecalis - An In Vitro Study

Background: Since the past decade, endodontics has been trending towards the utilization of natural products as root canal irrigants, medicaments, sealers, etc. This has numerous advantages of low cytotoxicity, cost, side effects, high biocompatibility with oral and periapical tissues, etc. The antimicrobial nature of neem and lemongrass separately, is well documented, but none have combined the effect of both these herbs when used together as a root canal irrigant.
Aim: The aim of this study was to evaluate the antimicrobial efficacy of a novel Neem-Lemongrass formulation as a root canal irrigant against E. faecalis.
Materials and Methods: Ethanolic extracts of Neem and Lemongrass were prepared separately and mixed in the ratio of 1:1. Antibacterial susceptibility testing was done using Agar well diffusion assay and compared with 3% sodium hypochlorite. Minimum inhibitory concentration (MIC), Minimum bactericidal concentration (MBC) values were determined by tube dilution test. Statistical analysis was performed using ANOVA.
Results: The largest zone of inhibition was shown by 3% NaOCl (20.1 ± 1.1mm). Zone of inhibition of 100µg Neem+Lemongrass irrigant (18.5 ± 1.9 mm) was comparable to that of 3% NaOCl (p<0.05). The MIC for the Neem-Lemongrass formulation was 12.3µg/ml and the MBC was 13.5µg/ml. The percentage inhibition of microbial growth of Neem-Lemongrass formulation at 50 (77.11%) and 100µg/ml (78.94%) concentrations was comparable to that of 3% NaOCl (84.89%).
Conclusion: Neem-Lemongrass extract showed good antibacterial activity against E.faecalis and hence, can be used as a potential root canal irrigant in future endodontic practice
