Association of 5 – HTTLPR (S allele) Polymorphisms with Depression, Schizophrenia and Anxiety

Abstract: Serotonin is considered one of the most necessary factors in current psychotherapy and a noteworthy amount of research is associated with this neurotransmitter in some way. Serotonin transporter (5-HTT) and its polymorphisms are implicit to be related with a numerous psychiatric disorders. In response to environmental stress, certain gene polymorphisms are increased the risk of psychiatric disorders by number of mechanisms includes: Regulation the metabolism of neurotransmitters and their receptors; Monitoring the synaptic factors and concentrations of such neuronal classes and directive of signaling transduction pathways in neurons. Several study suggested that the polymorphisms of the serotonin transporter gene (5-HTTLPR) have been strongly associated with different mental illness. Method: DNA was extracted from whole blood collected from 73 individual with mental disorders includes (37 depression , 16 schizophrenia and 20 anxiety) from Psychiatric Diseases Consultant /Al-Hussein General Hospital. In addition to 50 samples were collected as control group in a case-control study. The Variable Number-Tandem Repeats (VNTRs) polymorphism analysis for 5-HTTLPR gene was determined with PCR technique. Results: The 5-HTTLPR polymorphism was analyzed among both studied groups. The results revealed that there is no significant difference was detected between control and cases in (S/S) genotype and (S) allele of 5-HTTLPR polymorphism (P =0.27; P = 0.29) respectively. Whereas the heterozygote (L/S) genotype was strongly associated in case group (P = 0.04; OR= 1.86; 95% CI= 0.97-3.59). Conclusion: Our data suggested that there was no significant association in risk (S/S) 5-HTTLPR polymorphism genotype and (S) allele between a study groups. While Heterozygous (L/S) genotype carriers one copy of short allele were significantly greater occurrence in all case groups.
