Preparation, characterization and anticollagenase activity test of Alpinia zerumbet extract loaded NLC and cream formulation

Abstract: Purpose: This research aims to produce the anticolagenase activity in vitro from NLC (Nanostructured Lipid Carriers)of Alpinia zerumbetrhizome extractand stable cream. Methodology: The extract loaded NLC was prepared through high speed homogenization-ultrasonication method using solid lipids, liquid lipids, surfactants and extracts that have been made by means of multilevel extraction kinetically macerated with n–hexane, ethyl acetate and 96% ethanol. The cream was prepared with 3 formulaswith the variation of extracts and extract with the delivery system of NLC. The characterization, Scanning Electron Microscopy, and test on the anticolagenase activity were given for the extract loaded NLC produced. The physical quality of the cream was evaluated as well. Results:The results of the characterization of 96% ethanol extract loaded NLC showed the spherical-shaped particle size of 153.7nm, polydispersity index of 0.407 mV and zeta potential of -32.0mV.Meanwhile, the results from the anticolagenase activity (IC50) ofextract loaded NLC wasfound at 62.83µg/mL and the results of the cream evaluation based upon the statistic analysis of one-way ANOVA showed a significant difference in the globular size, viscosity and spreadability – except in pH.Then, it can be concluded that the best cream formula was found in the formula with the delivery system using the extract loaded NCL fulfilling the quality evaluation and potential to inhibit any anticolagenase activities to minimize the wrinkles.Value:From the research, it can be obtained the formula of nanoparticle cream with NLC delivery system from Alpinia zerumbet rhizome extract with anti-wrinkle activity.
