Abstract:

Purpose: The number of smokers is increasing in Indonesia. This causes many diseases caused by cigarette
smoke, one of which is lung cancer. Purple nutsedge (Cyperus rotundus l.) rhizomes are plants that have anticancer
effects. This study aims to determine the presence of cytotoxicity and antiproliferative effects of
methanol extracts of purple nutsedge (Cyperus rotundus l.) rhizomes in lung cancer line cells.
Methodology: This research used maceration method to extract the purple nutsedge rhizomes using purple
nutsedge rhizomes powder and methanol solvent. Extraction was tested at 600µl, 400µl, 200 µl, 100 µl, 50 µl
to determine the effect of cytotoxicity of compounds causing 50% cell death or IC50. Furthermore, IC50, 1/2
IC50, and 1/4 IC50 levels are used in the antiproliferative test to see the doubling time of the lung cancer line
cells. Cytotoxicity and antiproliferative tests used the MTT method to obtain purple formation crystals and
used an ELISA reader to obtain absorbance values.
Results: The result is a cytotoxicity and antiproliferative effect of methanol extract of purple nutsedge
rhizomes. IC50 of methanol extract of puzzle grass tubers was 235 µg/ml and doubling time at IC50 levels for 63
hours, 1/2 IC50 48 hours, and 1/4 IC50 30 hours, while in control for 20 hours.
Applications/Originality/Value: The methanol extract of purple nutsedge rhizomes can be said to have an anticancer
effect.
