Immunomolecular characterization of inflammation related caspases genes-5in patients with urinary tract infections causing Escherichia .coli

Abstract: In this study 180 urine samples, whole blood and serum were Collected from Patient suffering from symptoms referred as urinary tract infection. Samples were collected fromseven hospitals in Baghdad for six month.Depending on the characteristic examination, morphological of the colonies phenotypic and microscopic when grown in MacConkey agar and / or blood agar and HiCrome UTI agar, then finally by using vitek 2 a compact system . 56 (31.11%) urine samples gave positive bacterial growth, identified asE. coli. 17 isolates out of 56 positive cultures were identified as male samples (30.35%), while the rest was female samples 39 isolates were identified as E.coli (69.64%).Antibiotics sensitivity test was done by used 20 antibiotics; the isolates were multidrug resistance. Theisolates showed resistance as Follow: (96.4%) to Ampicillin, (89.2%) to Amoxicillin- clavulanic acid, (5.3%) to Amikacin, (48.2%) to Aztreonam, (89.2%) to Ceftazidime,(66%) to Ciprofloxacin (78.5%) to Cefepime, While all E.coli isolates showed sensistive to Imipenem and Meropenem. Detection of target genes by PCR detection was done by used conventional PCR for caspase-5 by using specific primer. The result for the casp-5 genes was positive for all isolates. Sequencing for 10 casp-5genes PCR product to detect variations in gene of Iraqi population and the results showed 10 samples from 10 had a genetic variation, heterozygous nucleotide. 56 patients and 24 healthy blood samples with age range (5 year - 74 year) and both gender, Enzyme-linked-immuno-sorbent-assay (ELISA) technique was used to estimate serum levels of IL-1ß and as immunological parameter for caspase-5 activation againstE.coli that caused urinary tract infection .The results of comparison between level of IL-1ß in patient infected with UTI and level of IL-1ß in healthy control was significant between them. p value 0.006.The comparison between the level of the IL-1ß in serum patient distributiongroups according to ages and the level of IL-1b in healthy control the results showed significant between the healthy control group and the A,B,C,E, and F group the p valu of them 0.04, 0.004,0.002,0.007 and 0.02respectively.(P< 0.05).andthe level of the IL-1ß in serum patient distributionaccording to gender and the level of IL-1ßin healthy control. The results showed significant between the healthy control and female patients and male patient, the p valu of them 0.01 and 0.04, (P< 0.05).
