Anti-candidal activity of ethyl acetate crude extract of endophytic fungus, Lasiodiplodia pseudotheobromae IBRL OS-64 against Candida albicans

Abstract: The present study was carried out to investigate the effect of ethyl acetate crude extract of endophytic fungus, Lasiodiplodia pseudotheobromae IBRL OS-64 against Candida albicans. The results showed that the fermentative broth extract possesses antiyeast activity whereas no activity observed for the fungal biomass extract. The ethyl acetate extract exhibited moderate anti-candidal activity against Candida albicans with inhibition zone was 11.7 ± 0.4 mm. The minimal inhibition concentration (MIC) and minimal yeastocidal concentration (MYC) values of the extract were 1.000 mg/mL and 2.000 mg/mL and showed yeastocidal activity at 2.000 mg/mL. The time-kill curve study demonstrated that the extract affected C. albicans cell growth with concentration and time dependent. Further detail of the effect of ethyl acetate extract on the C. albicans cell was observed with the aid of scanning electron microscope (SEM) and transmission electron microscope (TEM). The photomicrographs revealed that the fungal extract caused a severity of morphological deterioration to the treated cells. The data suggest that ethyl acetate extract of L. pseudotheobromae IBRL OS-64 showed significant anti-candidal activity against pathogenic yeast especially C. albicans.
