Deciphering The Genetic Alteration In Vegf Gene Family And The Putative Association With Oscc

Abstract: Vascular endothelial growth factor (VEGF) is an angiogenic factor which promotes the growth of new blood vessels. The levels of serum VEGF was found to be significantly higher in cancer patients compared to normal controls. The trend is found to rise with different clinical stages and involvement of lymph nodes. The present study follows an observational study design, which employs several computational tools for the identification of genetic alterations and variations in the selected dataset. The primary data set was obtained from cBioportal, which served as the source of information about the HNSCC patients. Whole genome sequences of the patients from different cohorts have been deposited for analysis. The information about the genetic alterations were provided in the form of Oncoprint data which was used for further assessments. The protein stability and pathogenicity was investigated by I-Mutant and PROVEAN tools. Among five crucial genes of the VEGF gene family, VEGFC showed a high frequency of gene alterations (3%). Most of these abnormalities were either gene amplifications or deep deletions. Gene expression patterns showed significant differences among grades of HNSC tumor. The survival probability of HNSC patients as depicted by Kaplan Meier plot showed that the differential gene expression pattern of the VEGFC gene was found to have an effect on the survival of patients producing a significant p value of 0.0029. A population wide screening of the novel variants identified in this study could aid in deriving an association between the genetic variants and HNSCC.
