Abstract:

Sulfonamide group is a magic group introduced as the main core for different bioactivities in drug industry. Pharmacologically and according to its substitution, sulfonamide derivatives could be divided into antibacterial sulfonamides and non-anti-bacterial sulfonamides, the later includes a wide range of medical uses such as anti diabetic agents ,carbonic anhydrase inhibitors , antihypertensive, antiviral, anti-impotence agent , high-ceiling diuretics, antiepileptic's and anticancer agents. These different pharmacological properties were found through the serial derivitization of the basic chemical nucleus of sulfanilamide, which indicate that these group has the ability to interacts with different targets. The anticancer activity of sulfonamides derivatives were produced by different mechanism of actions such inhibition of carbonic anhydrase (CA) enzymes, arresting the cell cycle in the G1 phase, and microtubules disruption. In this review, we try to summarizes and classify the different mechanisms by which sulfonamides derivatives produce there antitumor activity with illustration of the most important derivatives in each mechanism.
