Molecular characterizations of Echinococcus granulosus isolated from human and sheep in Euphrates region of Iraq .

Abstract:

The study included examine of 14 samples of hydatid cysts of Echinococcus granulosus (5 human samples and 9 sheep samples) During the study period from January 2019 - December 2019 . The study aimed to molecular identification of the common strains of Echinococcus granulosus in Euphrates provinces using PCR technique and study of the gene sequence of the strains and the drawing of the phylogenetic tree and comparing them with the strains recorded in the NCBI-BLAST to determine the most common strains in human in the Euphrates provinces. The sources of hydatid cysts samples in this study were from the livers and lungs. DNA was extracted from the germinal layer then the product was amplified by PCR technique by the DNA sequencing in gene 18S rRNA internal transcribed spacer 1 (ITS1) was targeted with a molecular weight of 579 base pairs( bp). The results were positive in seven samples and negative in seven samples .
The results in humans showed that the sheep strain (G1) is the common and responsible for human infections in the studied central Euphrates provinces, as the G1 strain was found in 4/5 of the samples, while the buffalo strain (G3) strain appeared in only one sample(1/5) with a percentage of 100% match with isolates and with total genetic variation (0.005-0.0250. %). And the sheep strain( G1) is common strain in sheep infections in the Middle Euphrates provinces (2/2).
