A study of the anticonvulsant activity of the stem bark of Persea americana

Abstract: The objective of the present study was to investigate the antiepileptic effect of the aqueous extract of Persea americana bark (AEPAB) and ethanolic extract of Persea americana bark (EEPAB). The antiepileptic activity was evaluated using Maximal Electro Shock (MES) and Pentylene Tetrazole Models (PTZ).Where in the first model of MES; convulsions were induced in wistar rats by putting them through electric shock (150mA, 0.2 sec) using ear clip electrodes. Three different dose levels (100, 200, and 400 mg/kg body weight) of AEPAB and EEPAB were used for the studies. Phenytoin was used as the standard drug. Various phases of convulsions were observed after MES i.e. Tonic flexion, tonic extensor and ratio of E/F (extensor to flexor) was observed. In PTZ model convulsions were induced in swiss albino mice of either sex weighing around 25-30g using PTZ at 60mg/kg body wt. and sodium valproate was used as the standard drug. EEPAB at dose of 200 and 400 mg/kg body wt decreased the convulsions ;( E/F) phase significantly .whereas the AEPAB decreased the duration of convulsion (Extensor to Flexion) ratio at a dose of 400mg/kg significantly. Similarly in PTZ induced seizures, EEPAB in doses of 100, 200, and 400 mg/kg delayed the onset of convulsions upto115.2± 6.973, 124.7±5.188, 153.0±4.899 seconds respectively. The results demonstrate that the EEPAB has a better potential for protection against convulsions in comparison with AEPAB.
