New developed method for determination and detecting of Sitagliptin Phosphate via the use of ISNAG –fluorimeter instrument

Abstract:
|
A new, simple, sensitive method has been developed for the determination of Sitagliptin Phosphate pure form and drugs by continuous flow injection analysis. The method was based on the reaction of Sitagliptin Phosphate (STP) with Phosphotungstic acid (PTA) in salt medium for the formation of a white precipitate, using homemade ISNAG –fluorimeter. Chemical and physical parameters were studied and optimized. The calibration graph was (0.5-6) mmol.L-1 and LOD was 20.933 ng/sample from the stepwise dilution for the minimum concentration in the linear dynamic ranged of the calibration graph .The correlation coefficient (r ) was 0.999. While percentage linearity (R2 %) 99.80 .The methods: developed method was applied successfully for the determination of STP in pharmaceutical tablets. A comparison was made between the newly developed method analysis with the classical method (UV- spectrophotometry at ? max =267 nm) using the standard addition method via the use of individual and paired t-test. It was noticed that there was a significant difference between two methods at 95 % confidence level.
