Solubility and Dissolution Rate Enhancement of Poorly Water Soluble Telmisartan by Melt Granulation Technique Using Soluplus and PEG8000 as Carrier

Abstract: Solubility can be defined as an important physicochemical factor influencing the drug’s absorption in addition to dissolution rates in the aqueous G.I.T fluid resulting in therapeutic effectiveness and inadequate bioavailability. The presented study attempted on enhancing the dissolution rates and the solubility regarding poorly water-soluble drug, Telmisartan (TEL), by using melt granulation technique with Soluplus® (graft copolymer of polyethylene glycol; polyvinyl carpolactam; and polyvinyl acetate) and Polyethylene glycol-8000(PEG-8000). Telmisartan has been prepared in ratios of 1:1, 1:2, and 1:3 of drug to polymer ratio (by weight) w/w. The formulations had been characterized with regard to their drug-polymer interactions, drug content studies, solubility parameters, and drug release studies with the use of FT-IR spectra. In addition, the selected formula (F-6) contains 1:3 ratio of the drug: soloplus specifying release of 99.49% in comparison to 35.82 % concerning pure TEL. At the same time, FT-IR studies revealed no change in the positions regarding TEL’s functional groups. The results obtained showed that soloplus is promising carrier might be used for improving the solubility related to poorly soluble TEL.
