Nimesulide is nonsteroidal antiinflammatory drug (NSAID). It’s permeability of the gel transdermally is non-effective therefore utilizing penetration enhancer could significantly improve the permeability. Many enhancers have been used for improving drugs penetration through the skin. However, most of them is toxic such as surfactant. Olive oil and almond oil alone and in combination with different concentration have been used in this study as safe enhancers to improve nimesulide permeability through transdermal route. The results show that Olive oil and Almond oil are safe and cause no irritation, as shown in the skin irritation test. In addition, they are both effective in increasing the permeability of the drug. It could be observed that the gel formulation of the drug without penetration enhancer had very small penetration, only 44% after 24 hours, while the percent’s penetration was reached to 59 ,81, 87, 93.3 and 94.5 when these oils were used separately. While when used together in combination, the percent penetration was increased to reach maximum 99.2 for the same period.
