Transferosomes: Effective Transdermal Drug Delivery Approach

Abstract:

Though conventional oral and parentral drug delivery system are used since long time but these both routes have certain limitations like hepatic first pass metabolism, invasive method, poor patient compliance, restricted patient activity etc. These limitations can be overcome by transdermal drug delivery system which provides advantages over these two drug delivery systems. Transdermal drug delivery system is non-invasive and patient compliance method that provide controlled drug release. But the main obstacle in the path of drug delivery through transdermal route is tough skin barrier which restricts the entry of majority of drug molecules especially those having high molecular weight and which are hydrophilic in nature. As a result, this method is useful for administering only few of the drugs. Various strategies has been studied and developed to overcome this problem and vesicular system is found to be the best option. In vesicular system, liposomes, niosomes, ethosomes, transferosomes etc. are there which can be used for this purpose but they all have certain limitations which do not allow them to suitable drug delivery system. Out of all these, transferosomes are found to be most effective for transdermal drug delivery as it has flexible and ultra-deformable character which help it to cross skin barrier easily as intact vesicle without any significant loss. Also transferosomes are capable to carry both hydrophilic and lipophilic drug moieties. This review covers the concept of transferosomes along with its mechanism of action, application and comparison with other approaches for transdermal drug delivery.
