The nuc gene as a potent source for Staphylococcus aureus to resist thermal processes

Abstract: Background: Staphylococcus aureus is distinguished from other Staphylococcus by possessing nuc gene, so nuc gene can be used as confirmed diagnosis for this bacterium. Amplification of nuc gene by using non comprehensive primer can't be given the required product thereby false result. The failure occurs due to differences in the sequences of the nuc gene forming nucleotides. So efficient primer should be chosen to amplify this gene. Methods:A number of 37 S. aureus isolates were isolated from food and clinical samples and identification with morphological and biochemical method , also by using confirmation tests included 16SrRNA gene which specific to S. aureus and nuc gene by PCR. Results:A Studied food samples are milk, cheese, yogurt, and cream , cheese with a percent (%33.3) was the most polluted . While in clinical samples( nasal swabs, urine swabs, and burn swabs), nose (%19) was the dominant with infected bacteria.The nuc gene amplification in all 37 S. aureus isolates. Conclusion: The nuc gene is a diagnostic feature of S. aureus and the variations that vary between strains of S. aureus should be taken into consideration when choose primers to amplify this gene. Thermonuclease encoded by nuc gene is a good weapon to resist thermal processing, thus contamination, especially with presence contaminated utensils and infected persons.
