Bioactive Fraction of Galangal Rhizome (Alpinia galanga): Determination of Acetoxychavicol Acetate Concentration By HPLC and Cytotoxyc Activity on T47D Cancer Cell Line

Abstract:

Background
Galangal Rhizome (Alpinia galanga) has the semi polar main content of 1'-acetoxychavicol acetate (ACA), which
is potential to be a cytotoxic agent in T47D cells. The present study has not evaluated the cytotoxic activity of
the galangal fraction against cancer cell line T47D more. This study aimed to determine the cytotoxic activity
of T47D cells and the concentration of ACA contained in ethanol extract, ethanol-water fraction, ethyl acetate
fraction, and n-hexane fraction of galangal rhizomes.
Materials and Methods
The ethanol extract of galangal rhizome was fractionated with liquid-liquid partition method. Cytotoxic activity
was carried out by MTT assay and ACA determination using the RP-HPLC method with the mobile phase of
acetonitrile: methanol (75:25), flow rate of 1 mL/minutes, and wavelength of 253 nm. The calibration curve of
ACA obtained was y = 2932.8x - 4848.4 with the correlation coefficient (r) = 0.999.
Results
The results showed the greater cytotoxic activity of ethyl acetate fraction compared to other samples with
IC50 values of 100.46 µg/mL and ACA content of 4.49 ± 0.09 mg/mL. Thus, the relation between cytotoxic
activities in the four samples is directly proportional to the ACA levels.
