Effect of Sea Cucumber Extract (Holothruidae sp.) on Pro-Inflammatory Cytokines (Interleukin-1ß, Interleukin-6) and Matrix Metalloproteinase-9 (MMP-9) of Periodontitis Wistar Rats (Induced Porphyromonas gingivalis)

Abstract: Introduction: Periodontitis is a multifactorial infectious disease that caused by complex bacteria species and the host response against these bacteria, that will cause periodontal structure destruction including alveolar bone and periodontal ligament. P. gingivalis is the main etiological agent in pathogenesis and inflammatory process of periodontal disease. Interleukin 6, interleukin 1ß play important role in developmental of chronic periodontitis. MMP 9 are the common MMP that are found in periodontal destruction. Sea cucumber is soft invertebrate from class Holothruidea. Sea cucumber has anti-cancer, antibacterial, antifungal, anti-inflammatory, anti-hypertention, antioxidant and antiviral properties Material and Methods: Sea cucumber (Holothrudidae sp.) extract was made by using ethanol solution and then rotavapor was used to get the extract. In this research, wistar rats weighted 200 – 250 mg were used, then Pohrphyromonas gingivalis was induced into rat’s gingiva. After 3 days, the rats’ gingiva were scaled and sea cucumber was given orally for P1 and P2 once a day for three days. After 3 days, K1 and P1 were sacrificed, the tissue was stored in -20°C and after 7 days, the K2 and P2 were also sacrificed and, IL-1ß, IL-6 and MMP-9 were measured using ELISA reader. Discussion: sea cucumber can reduce the expression level of IL-1ß, as pro-inflammatory cytokine and also has anti-proliferative effects on cancer cells. Sea cucumber extract may block prostaglandins and pro-inflammatory cytokines because of the PUFA content. Conclusion: sea cucumber extract (Holothruidae sp.) can suppress the pro-inflammatory cytokine such as IL-1ß, and MMP-9 that can play important role in periodontitis
