The Probable Synergistic Effect of Claramine with Five Flourouracil on Colon Cancer Cell Line HCT 116: In Vitro Study.

Colorectal cancer is third most common cancers and second cause of cancer-related death around the world. Five-Fluorouracil is widely used in colon cancer chemotherapy regimens. Unfortunately, failure of colorectal cancer chemotherapy had become more dominant due to the acquirement of 5-FU resistance by cancer cells. In this study we examined the cytotoxic activity of claramine on colon cancer cells as alone and in combination with 5FU.
Objectives: This study had been conducted to clarify the possible anti-proliferative potential of claramine on colon cancer cells and to illustrate a probable synergistic effect of claramine when it is given in combination with 5FU.
Materials and Methods: HCT 116 colon cancer cell line seeded on a 96-well plate was exposed to serial dilutions solutions of claramine, 5FU, and combination of both drugs. The plate then was incubated at 37ºC for 24hr. The cytotoxicity was measured by MTT assay.
Results: MTT viability assay showed that claramine significantly reduced viability % of treated HCT 116 cells compared with control HCT 116 cells. By using IC50 values of claramine and 5FU when used alone and in combination and applying them in Chou-Talalay's equation for combination index CI calculation, the CI was less than one meaning that there is synergism between 5FU and claramine when used in combination.
