The main aim of this study was to evaluate the antibacterial and antibiofilm action of chitosan nanoparticles
when used in combination with Metapex (A calcium hydroxide and iodoform based sealer which is most
commonly used in children) on the planktonic and biofilm states of the most commonly present gram negative
bacteria in the root canals of primary and permanent teeth- E.fecalis.
Methods and Results: E. Fecalis was subjected to investigations in both planktonic and biofilm states of the
bacteria, and the corresponding antibacterial and antibiofilm action was investigated. The test sample was
investigated using different concentrations of the nanoparticles and metapex in combination. The results
showed that chitosan nanoparticles exhibits abundant bacteriostatic effect on planktonic growth and biofilm
formation of E.fecalis in a concentration dependent manner but its bactericidal effect is less pronounced.
Polystyrene discs were coated with chitosan nanoparticles and metapex in various concentrations and was
found to reduce both early biofilm formation and late biofilm formation (7 days) which was confirmed by
fluorescence scanning electron microscopy.
Conclusion: This study demonstrated that chitosan nanoparticles when added to Metapex, a root canal
sealant has excellent bacrteriostatic effect against E.fecalis and that the action is dependent on the amount of
chitosan added. In addition, they have immediate and late antibiofilm action.
Significance of the study: Chitosan can have an effective impact against E.Fecalis bacteria when used along
with Metapex, commonly used root canal sealant in pediatric teeth.
