Cytotoxic study to evaluate the safe dose of mefenamic acid and aceclofenac for intracanal analgesia

Abstract: Mefenamic acid is a Non Steroidal Anti Inflammatory Drug (NSAID) belonging to an anthranilic acid group while Aceclofenac is an acetic acid derivative. Aceclofenac is a non selective cyclooxygenase inhibitor and Mefenamic acid is a potent cyclooxygenase inhibitor. Mefenamic acid is not being commonly used because of its gastrointestinal side effects like diarrhea, steatorrhoea and inflammation of the bowel. Aceclofenac has also shown gastro intestinal irritation. Looking at the brought forth side effects, it is beneficial to use Mefenamic acid and Aceclofenac as intracanal analgesics where its actions can be localized without having systemic side effects which is the core idea of this study. The cytotoxic effects of various concentrations of mefenamic acid and aceclofenac solutions were evaluated against brine shrimp nauplii at the end of 6, 12 and 24 hours and its lethal concentration (LD50) that killed 50% of the study population was calculated. The LD50 of Mefenamic acid solution for intracanal use was calculated to be 500µg/ml and LD50 of Aceclofenac could not be determined for the tested concentrations and was presumed to be greater than Mefenamic acid. This gives a wide safety window for use of these agents for intracanal analgesia.
