Using Alcoholic Extract of Curcuma longa L. Rhizomes as Staining of Bacterial Spores

Abstract: This study aims at using alcoholic extract of Curcuma longa L. rhizomes. The alcoholic extract has been prepared by using 20 gm. of dried powder that has extracted by Soxhlet extractor by using 200 m. of Ethanol “ethyl alcohol”, the concentration of Ethanol is 95% for 12 hours. The obtained results have been used in order to stain the bacterial spores that product Bacillus spp., the results display that staining of bacterial spores with brilliant yellow (Alcoholic Extract of Curcuma longa L. Rhizomes) is considered as a primary stain, but the vegetative cells appear pink/red of Safranin O as a secondary stain, bu using a method like Schaeffer-fulton method or Spore satin that by replacing Malachite green, which was responsible for the staining of bacterial spores with alcoholic extract of curcuma longa L. rhizomes.
