Evaluation of Ocular Retention and Bioavailability of Mucoadhesive Bromfenac Nanoparticles By Sustained Ophthalmic Delivery

Abstract: Delivery of drug to eye is eye through topical route of administration, however majority of drug is eliminated without absorption due to blinking of eye and nasolacrymal secretions. Ocular barriers will not allow the absorption of foreign materials, including therapeutic agent. The research work aim to formulate sustained release bromfenac mucoadhesive loaded nanoparticles as a novel drug delivery system. The formed nanoparticles through ionic interactions among chitosan and sodium alginate elicit significant penetration and prominent surface charge-dependent mucosal properties. The in vivo results indicates that no sign of irritation or damage to ocular tissues in the eyes of rabbits. In relation to the Megabrom (marketed formulation), the OBF shows significant (p<0.05) enhancement of AUC, AUMC, Cmax and elimination rate constant. The MRT of OBF was higher than that of standard Megabrom. Developing BF-loaded NPs using chitosan and sodium alginate in an effort to improve its ocular absorption and resulting high therapeutic efficacy. The pharmacokinetic parameters clearly evidenced that the prepared nanosuspensoions has high ocular retention, reducing frequency of administration, there by improving patient compliance and easily accessible to clinically applicability. This prepared & selected formulation, drug release is a viable alternative to conventional ophthalmic suspensions based on its drug release to elicit its therapeutic effect and improves patient compilance.
