TLR-4 and IFN-? levels in aborted women with IgM seropositive anti–toxoplasma antibodies

Abstract: Background: Toxoplasma gondii is a protozoan that can cross the placenta of the infected mother and causes severe fetal damage or abortion. The glycosylphosphatidylinositol (GPI) of T. gondii was demonstrated to trigger TLR4 signaling pathway and stimulate the production of interferon (IFN?) through MyD88 signaling that lead to formation of Th1 type response and activates lymphocyte cytotoxicity than cause termination of pregnancy. Aim: To determine the association of serum level of TLR4 and IFN ? with IgM seropositive anti –toxoplasma Abs in aborted women Materials and Methods: Fifty aborted women with IgM seropositive for Toxoplasma gondii and fifty aborted women with IgM seronegative for Toxoplasma gondii as controls were enrolled in the study. Three ml of blood collected in plain tube for separation of serum. All samples, were subjected for detection of serum level of TLR4 and IFN ? by ELISA. Results: Median serum level of IFN-? in patients was 377 pg/mL (range 32-796 pc/mL) compared to 69 pg. /ml (range 35-597 pc/ml) in controls with a highly significant difference. For sTLR-4 the median serum level in patients was 2.12 ng/ml (range 0.57-8 ng/ml) which significantly higher than that of controls (median 0.95 ng/ml, range 0.4- 6.9 ng/ml). Conclusion: Serum levels of soluble TLR-4 and IFN-? are elevated in women with with IgM seropositive anti –toxoplasma antibodies with no significant correlation between the two markers, and the sensitivity and specificity of the test at cut off value of IFN-? indicating a moderate discriminative value. While For sTLR-4 indicating good sensitivity and poor specificity.
