Detection of Theileriaannulata isolated from cattle using Real-Time PCR

Abstract:

This study aimed to molecular investigation the prevalence of Theileriaannulataparasite that isolated from blood
samples of cattle. A hundred of blood sampleswascollected randomanly from cattles ofthe middle area of Iraq
(Babylon province). Samples were submitted to blood DNA extraction assay. Nano drop spectrophotometer and
QRT-PCR with TaqMan methods were involved for the surface antigen Tams1detection of Theileriaannulata. Real-
Time PCR results showed only 16 samples were positive out of 50 blood samples with a percentage 32%. The
threshold cycle (CT value) of these positive samples were ranged from CT: 24.29 to CT: 35.69. The Real-Time
PCRefficiency was validated using standard curve method, and the efficiently was approximately (99.9%).Surprisingly,
the overall results from current study showed there were no association between ages, gender. Infectionsdistribution
between (1-12month) (28.57%), 1-2 year (31.57%) and 3-10 year (29.4%).Additionally, the ratio of the infected
females at age of 1-2yearswere twice more than the ratio of infected males.
