Synthesis of Nanoencapsulation of Alpinia Galangal Extract Using Chitosan-Alginate and Cytotoxic Activity on MCF-7 Cell Lines

Previous research stated that Alpinia sp. of Indonesian local market has Trans p-coumaryl diacetate as anticancer. The
research was aimed to formulate nanoencapsulation of Alpinia sp. extract and the cytotoxic activity on MCF-7 cell
lines. The formulation of nanoencapsulation used chitosan-alginate and 1-ethyl-3-(3-dimethyl aminopropyl)
carbondimides (10:1:1) as matrix. Nanoencapsulation formed was characterized by spectrophotometry UV-Vis and
particle size analyzer (PSA). Drug loading (DL) and encapsulation efficiency (EE) were determined by RP-HPLC
usingmethanol: water (75:25 v/v) and were monitored at the wave length of 253 nm. The result showed that the
formation of nanoencapsulation was due to the particle size measured value of 264.3 ± 24.39 nm and zeta potential
value of 72.33 ± 2.50 mV. The drug loading of nanoencapsulation was obtained at 0.638 ± 0.019 % with the EE value
of 46.819 ± 1.381%. The nanoencapsulation of Alpinia sp. extract had cytotoxic activity on MCF-7 cell lines, IC50 value
was at 168.146µg/mL equivalent to 1.075 µg/mL of coumaryl diacetate. In view of the cytotoxic activity, it could be
concluded that trans p-coumaryl diacetate in nanoencapsulation form has increased the activity.
