SYNTHESIS AND CHARACTERIZATION OF ISONIAZID INCORPORATED THIOFIBRATES

Abstract: In the present study two novel thiofibrates bearing 1, 3, 4-oxadizole and 1,2,4-triazole are prepared from isonicotinic acid hydrazide (1). The compound (1) is on reaction with carbon disulfide in the presence of potassium hydroxide gave 5-(pyridin-4-yl)-1,3,4-oxadiazol-2-thiol (2). This on treating with ethyl-2-bromo isobutyrate in presence of anhydrous potassium carbonate in DMF yielded compound ethyl 2-methyl-2-[{5-(pyridin-4-yl)-1,3,4-oxadiazol-2- yl}sulfanyl] propanoate(3). Compound 1 on further reaction with phenyl isothiocyanate in presence of sodium hydroxide forms 4-phenyl-5-(pyridin-4-yl)-4H-1,2,4-triazol-3-thiol) (4). This when treated with ethyl-2-bromo isobutyrate under the similar conditions in obtaining compound (3) gave ethyl-2-methyl-2-{[4-phenyl-5-(pyridin-4-yl)- 4H-1,2,4-triazol-3-yl] sulfanylpropanoate (5).
