Investigation of USPION genotoxicity on Cytokinesis Blocked Micronuclei

Abstract: Micronuclei (Mn) are structures that arise from acentric chromosome fragments or complete chromosomes that failed to stick to mitotic spindle during cytokinesis and are rejected from the daughter nuclei into the cytoplasm. In this experiment, we tested human lymphoblastoid-B cells were treated with different ultrafine superparamagnetic iron oxide nanoparticles (USPION) and performing the micronucleus assay from genotoxic and cytotoxic effects by measuring the frequency of binuclear Bn and Mn cells in a cell population analysed. The results showed that treating with 50nm USPION was more effective than 600nm nanoparticles as it presented significant decline in %BN curve with a sudden increase for %Mn/BN curve. The examined human cells appeared some resistance to the external changes, at the first time, and then looked to be exhausted to face the additional doses.
