The effect of Hydro alcoholic Olive Leaf Extract on Vascular Endothelial Growth Factor GeneExpression Level in Ischemic Muscle in Hind Limb Ischemic Rat Model

Abstract:

Angiogenesis is said to be the formation of new capillaries from existing vascular network. One of the effective factors
on angiogenesis is endothelial growth factor (VEGF), which with its receiver plays an important role in angiogenesis.
Olive leaf through combination of polyphenols and flavonoids in itself has anti-oxidant and anti-inflammatory effects as
well as the direct and indirect effects on VEGF gene expression. In this interventional study, male Wistar rats were
divided into 4 groups. In ischemia groups, gastrocnemius artery was blocked after anesthesia with ketamine. For 21
days, the control groups (sham-ischemia) and the treatment groups (sham- ischemia), took normal saline and olive
leaf extract orally (300 mg/kg), respectively. After anesthesia, the rats Gastrocnemius muscle were separated from the
body. VEGF gene expression in muscle tissue was measured by Real Time PCR method. In this study, the mean of
VEGF gene expression in the treatment ischemia group (300 mg/kg) significantly decreased as compared to that of the
control ischemia group (P?.007). VEGF gene expression was more decreased in the treatment sham group than in the
ischemia control group, and this reduction was significant (P?.045). no significant difference was observed in gene
expression between the two treatment sham and ischemia groups (P?.467). According to the results, olive leaf
extract reduces VEGF gene expression in the ischemic muscle.
