Neuro protective activity of Sargassum Wightii against neurotoxicity induced cell death on an In Vitro human neuro blastoma model

Abstract: Parkinson’s and Alzheimer’s diseases are the result of the progressive dysfunction involving the central nervous system. Although, these diseases such as PD and AD have been widely studied over the past decades. Efficient treatments for these troubling diseases are not yet determined and only some therapies only for the time being improvement in certain symptoms. The objective of the current work was to investigate the probable neuro protective activity of S.wightii after the induction of neurotoxin 1-methyl-4-phenlypyridinium (MPP+) in SH-SY5Y human neuroblastoma cells. Neuro protective activity of S.wightii extract were evaluated for cell viability analysis, and IC50 determination, and intracellular reactive oxygen species, and acetylcholine esterase activity. The exposure of SH-SY5Y cells to MPP+ along with extract reduces the cell death up to 80 % for varying concentration in cell viability analysis. In the ROS assay, there was an increase in the free radical scavenging activity with the extract’s rising concentration. It indicates that the S.wightii extract reduces the oxidative stress produced by the neurotoxin. The acetylcholine esterase inhibitory activity also increased while compared with standard drug Donepezil. These observed results suggested that the methanolic extract of S.wightii can be a resource for novel drug for neurodegenerative diseases.
