Molecular screening of E.colibiofilm and anantibiofilm activity ofCapparisspinosa L extracts.

Abstract: Escherichia coli is one of the main causative agents of diarrhea and other intestinal diseases which due to their virulently and colonization ability playing the main role in the development of virulent cariogenic biofilms. Different cell surface and structure molecules were a reason for the development and composition of the biofilm formation ofEscherichiacoli. This study designedto identify the E coli strains isolated from diarrheal patients using specific and virulent gene and use of Capparis spinosa extract in inhibition of bacterial biofilm formation ability Three hundred eleven stool samples were collecting from children with diarrhea attended to Hilla Teaching and Al-Hashmiya hospitals from October 2017 – April 2018. Diagnosis of E. coli isolates depends on standard laboratoryprotocols and molecular detection method using specific primers based on16srRNA gene as a genetic marker for confirmed isolationEscherichiacoliby PCR. Only 45 isolates were showed positive results for these genes. Investigation of isolates activity for biofilm formation were achieved byTissue culture plate (TCP) assay. From50isolates of E. coliwere collected from children with diarrhea only 45 of these isolates gives positive results for 16sRNA gene.E. coliisolates were biofilm former, high, moderate biofilm formation mode was account for (70%) and (30%) respectively while there are no isolates expresednon biofilm formation. Regardingthe effect of flower and leaf of C. spinosa on biofilm formation, the result of this study demonstrates flower hotaqueousextract effectively reduces the biofilm formation to 33% moderate ability, 77% weak and no strength. While leaf extract at different concentrations not prevents the biofilm formation by E.coli. Molecular technique by using specific primers based on 16srRNA intergenic spacer gene as a genetic marker give accurate identification for E.coli. Capparis spinosa, extracts have an effect onE. coli biofilm formation.
