Development and validation of stability indicating HPTLC method for determination of Rutin and Quercitin

Abstract: A new simple and selective stability- indicating high performance thin layer chromatographic (HPTLC) method has been developed and validated for simultaneous estimation of Rutin and Quercitin. The chromatographic development was carried out on TLC plates Precoated with silica gel 60 F254 using mixture of Ethyl acetate: Toluene: Methanol: Formic acid (5:3:2:0.5) v/v as mobile phase. Both the markers showed considerable absorbance at 255 nm. Hence it was chosen as detection wavelength. Stress testing of Rutin and Quercitin was carried out according to the international conference of harmonization (ICH) guideline Q1A (R2). The markers were subjected to acid, base, neutral hydrolysis, oxidation, thermal degradation and photolysis. . The system showed a peak for Rutin and Quercitin at Rf value of 0.24 ± 0.02 and 0.68 ± 0.03, respectively. The method was successfully validated according to ICH guidelines Q2R1. The data of linear regression analysis indicated a good linear relationship over the range of 100–500 ng/band concentrations with correlation coefficient value greater than 0.995. The accuracy of the method was established based on the recovery studies. Under various stressed conditions, Rutin and Quercitin do not showed any degradation product under alkali, acidic, oxidative and neutral hydrolytic condition.
