Stability Indicating HPTLC Method for Determination of Metoprolol Succinate and Olmesartan Medoxomil in Bulk and in Formulation

Abstract:

An approach for the stress degradation was successfully applied for the development of stability indicating HPTLC method for the determination of Metoprolol succinate and Olmesartan medoxomil on the plates precoated with silica gel 60 F254. The mobile phase used was containing n- butanol and Methanol (6:3 v/v). Both the drug showed considerable absorbance at 216 nm. Development and Validation was carried out according to the international conference of harmonization (ICH) guideline Q2 (R1). The system showed a peak for Metoprolol succinate and Olmesartan medoxomil at Rf value of 0.23 ± 0.02 and 0.65 ± 0.05, respectively. The data of linear regression analysis indicated a good linear relationship over the range of 250–2500 ng/band and 100- 1000 ng/band for Metoprolol succinate and Olmesartan medoxomil, respectively with correlation coefficient value greater than 0.992. The accuracy of the method was established based on the recovery studies. The LOD and LOQ were 46.08, 11.51 ng/band and 139.64, 34.90 ng/ band for Metoprolol succinate and Olmesartan medoxomil, respectively
