Abstract:

The objective of this study was to evaluate the combination of pH-dependent and time-dependent
polymers by a statistical full factorial design. Eudragit S100 was used as pH-dependent polymers and
Eudragit RS100 and Eudragit RL 100 were used as a time-dependent polymer. Ornidazole was taken as a
model drug. Dissolution studies of tablets in the media with different pH (1.2, 7.4 and 6.8) showed that
drug release in colon could be controlled by addition of Eudragit RS and Eudragit RL100. The various
batches were prepared by using 32 full Factorial design in which X1 was the ratio of Eudragit RS 100 to
the RL 100 (25%, 50%, 75%) and X2 was the coating level of Eudragit S100 ( 0%, 5%, 10%). Two response
variables were amount of drug release up to 5 hr. and amount of drug release up to 12 hr. An optimized
formulation was found to be the one containing 5% coating of Eudragit RS:Eudragit RL (1:1) and
10% coating level of Eudragit S100.The experimental values of response variables highly agreed with
predicted values. From results obtained in the present study, it was concluded that an optimized formulation
successfully deliver the maximum amount of drug in intact form to the colon.
