Chromatographic simultaneous determination of Cefixime trihydrate and Linezolid in combined tablet dosage form

Abstract:
|
A simple, precise, accurate and sensitive Reverse Phase High Performance Liquid chromatographic (RP-HPLC) method and High Performance Thin Layer Chromatographic (HPTLC) method developed for simultaneous estimation of Cefixime trihydrate and Linezolid in combined tablet dosage form. Drugs were resolved on a C-18 columns (250mm ×4.6mm i.d. with particle size of 5µm) utilizing mobile phase of Acetonitrile:equal volume of methanol and H2O with 0.2%, 5ml Triethyl amine(15:85) pH 6.5 adjusted with 0.4% Ortho Phosphoric Acid with flow rate0.96 ml/min in HPLC. In case of HPTLC mobile phase isToluene : Methanol : TEA :NH3 (5:4:0.7:0.3 v/v).The regression analysis data for the calibration plot showed good linear relationship in the concentration range of 30-70 µg/ml (R2 = 0.9971) for Cefiximetrihydrate and 90-210 µg/ml (R2 = 0.9999) for Linezolid by HPLC method and 300-700 ng/band(R2 = 0.9971) for Cefixime trihydrate and 900-2100 µg/ml (R2 = 0.995) for Linezolid by HPTLC.The average percentage recovery of Cefiximetrihydrate and Linezolid combination was found to be 99.28% and 99.58% respectively by HPLC and 99.10% and 99.76% respectively by HPTLC.Statistical analysis proves that the method is precise, reproducible and selective for the simultaneous determination ofCefixime trihydrate and Linezolid in their combined tablet dosage form.
