Preparation, characterization and cytotoxic effects of nanoliposomes containing umbelliprenin

Abstract: Drug discovery from plant sources has drawn much attention these years. Natural products are a good source for finding new bioactive compounds. Umbelliprenin (UMB), derivated from Ferula species, has been used and investigated for its pharmacological activities. This natural compound shows various biological effects and one of the most important ones is cytotoxicity and cytostatic activity against tumoral cells. In the present study, to overcome the problem of insufficient water solubility of UMB, after synthesis of this compound with a chemical interaction between 7-hydroxycoumarin and trans-trans-farnesyl bromide, liposomes were selected as a carrier of UMB. The physical characteristics, stability and incorporation of UMB were determined. Then the cytotoxic activity of liposomal UMB against human tumoral cell line A375 was compared to the free form in DMSO. Mean size of liposomes was around 150 nm and they were stable during storage at 4 °C up to two months. The cytotoxicity results indicated that the liposomal UMB had better cytotoxic effects on melanoma cell line compared to the solution in DMSO. This effect was time- and concentration-dependent. Therefore, liposomal carriers showed the characteristics of a perfect system for delivery of UMB and in the future, it may be used in tumor chemotherapy.
