Antiproliferative, cytotoxic and anti-angiogenic potential of citral against U87MG cells

Abstract: Objective Glioblastoma Multiforme is rapidly growing malignant brain tumor which remains incurable in spite of current treatments like external beam radiation, surgery and chemotherapy. Citral, a key component of essential oils extracted from several plants, was reported to be cytotoxic. In current study, we evaluated anticancer effect of Citral on U87MG glioblastoma cells and heterotropic xenograft model in mice. Methods U87MG cells were treated with different concentration of citral (25, 50, 75 and 100 µg/ml) and cell count and cell viability were determined using Trypan Blue dye exclusion assay. Anti-angiogenic effect of citral was evaluated by Chorioallantoic Membrane assay. Effect on total vessel length and number of branching point were evaluated by ImageJ and Angiotool software. U87MG cells were implanted under transparent kidney capsule of Swiss Albino mice and treated with three different dose (100, 200 and 400 mg/kg) of citral. Tumor growth was assessed on 6th day of implantation. Results U87MG cell treated with citral showed significant concentration dependent decrease in cell count (4.81 ± 1.13 for 100 µg/ml versus 13.74±1.04 in control) and cell viability (Decreased up to 38%). It significantly decreased total vessel length and number of branching point in CAM assay. Tumor volume was found to be decreased by 87% in highest dose when compared with control in Subrenal Capsule Assay. Conclusion Citral was found to be antiproliferative and cytotoxic in U87MG cells. It showed anti-angiogenic potential in CAM assay and anti cancer effect in heterotropic xenograft model.
