Stability Indicating Method Development And Validation For The Simultaneous Estimation Of Orientin, Vitexin And Luteolin In Ecbolium viride (Forssk) Alston

Abstract: Flavonoids are a group of plant metabolites thought to provide health benefits through cell signalling pathways and antioxidant effects. The development of quality-control methods for determination of the major flavonoids in Ecbolium viride (Forssk) Alston is therefore essential for identification and evaluation of the quality of this medicinal plant. A method for simultaneous estimation of three flavonoids by reverse-phase high performance liquid chromatography (HPLC) was developed and validated. The flavonoids were analysed on a kinetex C18 100 A0 C18 column (250 mm × 4.6 mm, 5 µ particle size); using a mobile phase, consisted of binary mixture of Water pH adjusted to 3.5 with formic acid: Methanol (60:40); under the following conditions: detecting wavelength, 350 nm; flow rate, 1.0 ml/min. The HPLC system was operated at ambient temperature (30±2 0C) and the optimised method was validated. The calibration curves were linear over the range of 10- 80 µg/mL, 10-80 µg/mL and 5-35 µg/mL for orientin, vitexin and luteolin respectively (r > 0.999). The assay recoveries for three flavonoids were above 99.3%. (% RSD<1). The precision results (R.S.D.) for orientin, vitexin and luteolin were less than 1. The LOD and LOQ for orientin is 2.79 µg/mL and 9.30, for vitexin 1.97 µg/mL and 6.57 µg/mL and for luteolin 0.58 µg/mL and 1.93 µg/mL respectively. This method demonstrated good reliability and sensitivity, and can be conveniently used for the quantification of orientin, vitexin and luteolin in herb of Ecbolium viride
