Formulation and Evaluation of Colon Targeted Pulsincap of Lansoprazole – A Chronotherapeutic Approach

Abstract:

The aim of the present study was to develop colon targeted drug delivery system for Lansoprazole. Microspheres were prepared by ionotropic gelation method using sodium alginate and guar gum. The physicochemical characteristics of Lansoprazole microspheres were evaluated. Best formulation was selected for further preparation of hard gel capsules. The hydrogel plugs were punched using HPMC. Capsule bodies were made insoluble by formaldehyde treatment. The ‘pulsincap’ was evaluated for in vitro dissolution studies and drug release kinetics. Lansoprazole microspheres were spherical in nature and were confirmed by SEM studies. A maximum of 90% drug entrapment efficiency was obtained in Lansoprazole microspheres. The in vitro studies of Lansoprazole microspheres showed sustained action. The in vitro dissolution profile of Lansoprazole from modified pulsincap was in proportion to the concentration and viscosity of the polymer used in the hydrogel plug. The regression co-efficient values indicated that the release data fitted zero order kinetics.
