Development of Fast Dissolving Tablets of Nebivolol Hydrochloride by Solid Dispersion Method

Abstract:

Nebivolol hydrochloride is an anti-hypertensive drug used for the treatment of hypertension. The drug is classified under BSC class II, it has poor water solubility and bioavailability (<12%). The present approach of solid dispersion was used to enhance the solubility and subsequent dissolution of the drug. The solid dispersion was prepared by kneading and fusion method where mannitol was used as carrier in the ratio of 1:1, 1:2 and 1:3. The drug polymer interactions were studied using IR spectroscopy and DSC. The solid dispersion prepared using fusion method in the ratio 1:3 was found to be best among the other combinations. It showed in-vitro drug release of more than 50% in 60 minutes. Fast dissolving tablets were prepared using crosspovidone as a super-disintegrant, where the drug release in tablets was 90% in 30 min.
