A comparative study of adaptogenic effect of Withania somnifera with Mifepristone (a GR antagonist) on chronic stress - induced perturbations in glucose metabolism

Abstract:
|
Objective The study investigated the influence of crude extracts of ashwagandha and a novel fraction on glucose metabolism and comparing with standard glucocorticoid receptor antagonist, Mifepristone.
Methodology Adult male rats were randomly segregated into 6 groups (n=5), viz. controls, vehicle controls, stress, stress +mifepristone , stress +ethanolic, stress +chloroform extract and stress + fraction of ethanolic extract. They were exposed to restraint and forced swimming for 4 weeks.
Key findingsA significant increase (p <0.05, ANOVA) in adrenal 3ß-HSDH and 11ß-HSDH, adrenal gland weight, corticosterone and insulin levels, blood glucose level, activities of hepatic gluconeogenic enzymes (PEPCK and G6Pase), GGT and aminotransferases (GPT and GOT), and lactate and pyruvate concentrations, significant reduction (p <0.05, ANOVA) in hepatic and calf muscle glycogen and activities of hepatic PK, PDH, and G6PDH, GST and GR in chronically stressed rats suggested dysregulated glucose metabolismwere normalized by crude extracts and isolated fraction which was similar to mifepristone treatment.
Conclusions The results indicated stress-induced hyperglycemia accompanied by insulin resistance and glucose intolerance. Stressed rats receiving prior treatment of crude extracts or isolated fraction or mifepristone did not show these metabolic alterations despite undergoing stress. Further, ashwagandha suppressed stress-induced increase in corticosterone secretion suggesting its action through HPA axis.
