RP-HPLC method for the simultaneous determination of mefenamic acid and drotaverine hydrochloride in tablet dosage form.

Abstract:

A simple, specific, accurate and precise reverse phase high performance liquid chromatographic
method was developed for the simultaneous estimation of mefenamic acid and drotaverine
hydrochloride in tablet dosage form. A hypersil ODS-C18, 5 µm column 250 mm × 4.6
mm in isocratic mode, with mobile phase containing water, acetonitrile and methanol in proportion
of 30:35:35 v/v/v, pH 2.25 adjusted with orthophosphoric acid were used. The flow
rate was 1 ml/min, and effluent was monitored at 226 nm. The retention time of mefenamic
acid and drotaverine hydrochloride were 11.41± 0.07 and 14.34 ± 0.05 min respectively, and
the resolution factor was 3.0. Linearity range for mefenamic acid and drotaverine hydrochloride
was 8-24 µg/ml. The limit of detection and limit of quantification for mefenamic acid
was found to be 0.259 and 0.785 µg/ml respectively; and for drotaverine hydrochloride, 0.128
and 0.387µg/ml respectively. The proposed method is accurate, precise, specific and rapid for
simultaneous estimation of mefenamic acid and drotaverine hydrochloride in tablet dosage
form.
