Role of Alpha-7 Nicotinic Acetylcholine Receptors in Arthritis

Abstract: Alpha 7 nicotinic acetylcholine receptors (a7 nAChRs) are widely expressed in the nervous system, and also found in non neuronal cells like macrophages, lymphocytes, monocytes, endothelial cells, neutrophills, dendritic cells and tissues. Activation of a7 nAChRs inhibits overproduction of pro-inflammatory cytokines. Present study was carried out to find the possible role of a7 nAChRs in Complete Freund’s Adjuvant (CFA) induced arthritis. Arthritis was induced in adult female Albino Wistar rats by injecting CFA. 1,1-dimethyl-4-phenylpiperazinium (DMPP) was used as a non selective nAChR agonist and a-Bungarotoxin (a-BgTX) as a selective a7 nAChR antagonist. The effects of drugs were evaluated by various physical, biochemical and histopathological parameters. DMPP produced marked anti arthritic effect through a7 nAChRs which was antagonized by a-BgTX suggesting the possible role of a7 nAChRs in inflammatory pathways involved in rheumatoid arthritis.
