In this article, an overview of hydrophobic interaction chromatography (HIC) of proteins
is given. After a brief introduction of protein hydrophobicity and hydrophobic interactions,
we present the stationary phases that are used for this technique, salts used, comparison between
reversed phase HPLC and HIC, factors affecting HIC and experimental design. Selected
examples of protein purification by HIC are also presented.
