Formulation and Evaluation of Sustained Release Matrix Tablets of Glipizide using Optimized Batch of Solid Dispersion

Abstract:

The goal of the study is to develop once daily sustained release matrix tablet of Glipizide by applying solid dispersion
technique for improving solubility. The matrix tablets of glipizide solid dispersion granules were prepared by wet
granulation method using HPMC K 100 M as hydrophilic polymer and Eudragit RSPO as the hydrophobic polymer.
Preformulation studies were carried out. The matrix tablets were evaluated for their physicochemical properties, in vitro drug
release. To achieve complete drug release, various batches of solid dispersion of glipizide using water soluble carriers such
as PEG (PEG 4000 as X1 and PEG 6000 as X2), were prepared as per the design expert (8.0.5) to optimize the drug release
profile using response surface methodology (Face centered central composite design). DSC and FTIR were used to
characterize the solid dispersions. No chemical interaction was found between Glipizide and polymers. Dissolution profile of
matrix tablets containing solid dispersion has shown increase in the release rate compared to matrix tablets alone. From this
study, it was clarified that solid dispersion granules was one of the promising sustained release system applying solid
dispersion technique for the poorly water soluble drug.
