Development of Taste Masked Rapid Dissolving Films of Levocetirizine by Using Natural Polysaccharides as a Film Former

Abstract: Levocetirizine is a third-generation non-sedative antihistamine, works by blocking histamine receptors. It does not prevent the actual release of histamine from mast cells, but prevents it binding to its receptors. Levocetirizine prevents the release of other allergic chemicals produced in response to allergens and increased blood supply to the area, and provides relief from the typical symptoms of hay fever. This work aimed to develop rapid dissolving films of Levocetirizine for the treatment of allergic conditions. Rapid dissolving films were prepared by solvent casting method using pullulan PI 20 as film former and PG and tween 80 as plasticizers. Bitterness of Levocetirizine was masked by forming inclusion complex with HP?-CD. The complex was evaluated by XRD, DSC and FT-IR. Optimized films were evaluated for mechanical properties, surface pH and dissolution characteristics. The combination of pullulan PI 20, PG:tween 80 and xanthan gum exhibited excellent mechanical properties (Tensile strength 6.18 N/mm2), drug content (96.4 %) and disintegrating time (16 sec). The research work done here suggested the exciting possibility of Levocetirizine as rapid dissolving Film.
