Synthesis, Characeterization and in vitro Pharmacological Activity of novel 3-(4-Fluorophenyl)-1- isonicotinoyl-5- (substituted phenyl)-formazans.

Abstract: In the present investigation, a series of 3-(4-fluorophenyl)-1-isonicotinoyl-5- (substituted phenyl)-formazans (4a-j) were synthesized by condensation of schiff base (3) and diazonium salt of various substituted aromatic amines (3a-j). The intermediate schiff base (3) was itself synthesized by condensation of isonicotinic acid hydrazide with pfluorobenzaldehyde (2). The structures of the compound have been confirmed by elemental analysis and spectral analysis. Newly synthesized compounds were tested for their in vitro anti-tubercular activity against Mycobacterium tuberculosis H37Rv using the BACTEC 460 radiometric system. Among the synthesized compounds, compound 5-(2,6-dichlorophenyl)- 3-(4-fluorophenyl)-1-isonicotinoylformazan (5m) and 3-(4-fluorophenyl)-1-isonicotinoyl-5- (4-sulfamoylphenyl) formazan (5r) were found to be more active agent against M. tuberculosis H37Rv with minimum inhibitory concentration of 0.0043 µM and 0.0040 µM.
