Acute and Sub acute toxicity study of Siddha herbal formulation Athimathura Chooranam in rat

Abstract:

Athimathura chooranam is a Siddha formulation indicated for the treatment of Respiratory disease mainly Kanam (Bronchial Asthma). Safety profile for this formulation has been not done yet, hence in this present study Acute and Sub acute toxicity study has been evaluated. OECD guidelines 425 and 407 were followed for the toxicity studies. Acute toxicity study was evaluated 2000 mg/kg as limit test in Wistar strain albino rats. Test formulations were administered to overnight fasted animals and parameters like body weight, behavioral changes, and mortality were assessed for 14 days. Likewise Sub acute study was evaluated using both Male and Female Wistar albino rats in three groups separately. The animals received two doses herbal product (200 mg/kg/day and 400 mg/kg/day) and 5ml of saline as control for a period of 28 days. Hematological and biochemical parameters were assessed. Results showed no significant changes in terms of behavioral changes, mortality, and body weight. On 28th day of experiment, blood sampling of animals was done for hematological and biochemical analysis i.e. liver and renal function parameters, lipid profile and then sacrificed for histopathological examination of liver, Heart and kidney.
