Effect Of Extract & Fractions of Fruits of Terminalia Chebula on Chemical Induced Memory Impairments in Rats by Different Models

Abstract: Aim- The aim of the present investigation is to determine effect of extract & fractions of fruits of terminalia chebula on chemical induced memory impairments in rats by different models. Material & Methods- The dried fruits of Terminalia chebula were extracted by successive solvent extraction method with the help of soxhlet apparatus. The plant material first extracted with petroleum ether then chloroform, acetone, methanol and ethanol. During first three trial rat was placed in the light chamber, and allowed them to explore in both the chambers, and the connection between two chambers was kept open during this time. In the 4th trial, as soon as the rat entered into the dark chamber, connection door was closed and a foot shock (2.5 Ma, for 1 seconds) was given to the rats and the animal was kept there for additional 10 seconds. Transfer latency (TL) was taken as the time taken by animal to move into one of the covered arm with all its four legs. TL was recorded on the first day. Hole-board apparatus was made up of a square wooden box (50 x 50 x 50 cm) with four holes (3 cm diameter) at each corner. Results- The % yield of ethanolic, methanolic, petroleum ether, chloroform and acetone extracts of dried fruits of Terminalia chebula was found 16.5, 7.4, 2.81, 9.2 and 5.8 % respectively. In a scopolamine treated group animals the % avoidance response decreased significantly to the extent of 26.33 % in acquisition trial as compared to normal vehicle control animals 55.83%, thus suggesting marked impairment of memory. T. chebula Ethanol extract (TC-ETOH) treated groups animals showed significant decrease 36.33 seconds in transfer latency (s) of acquisition trial as compared to disease control animals 53.67 seconds, thus suggested marked improvement in memory. Conclusion- Bioactive fractions ethylacetate, butanol and water soluble fractions isolated from ethanolic extract of T. chebula fruit were experimentally evaluated on exteroceptive or interoceptive learning memory paradigms in rats which are associated with complex behavioral, neurophysiological, neuroendocrinal and neurochemical changes.
