Simultaneous Hptlc Method Development For Gallic Acid, Oleanolic Acid And E-Guggulosterone

Abstract: A new simple, precise, accurate and validated HPTLC method was developed for simultaneous determination of Gallic acid, Oleanolic acid and E-Guggulosterone. The chromatographic condition for separation of these three phytoconstituents was performed over pre coated TLC Plates (60 F 254 10 cm x 10 cm) using Camag 100 µl HPTLC syringe, Automated TLC sample applicator Linomat V with Nitrogen as inert gas and WinCATs software. Among the different combinations of mobile phases used, best separation was achieved in Toluene: Ethyl acetate: Formic acid (7:?6:?1, v/v/v). Densitometric scanning of the plates directly at 270 nm was used for analysis of E-guggulosterone and Gallic acid, while as for analysis of Oleanolic acid; plates were scanned at 368 nm after spraying with methanolic-sulphuric acid reagent. The retardation factor (Rf) value of E-guggulosterone, Gallic acid and Oleanolic acid was 0.68, 0.32 and 0.8 respectively. Validation of this developed method has been performed to demonstrate Precision, Linearity, Accuracy, Robustness, LOD and LOQ. Applicability of this validated method was carried out in lab scaled formulated dosage form and determine % Assay of these three phytoconstituents. Such type of chromatographic fingerprint analysis may useful tool for quantification of Phytoconstituents in polyherbal formulation in future.
